PART 1: INTRODUCTION

1. - Background Information

Please find attached the Request for Proposals documentation with specifications and the CFI guidelines pricing format to be completed and returned with your proposal.

1. - Statement of Requirements

1.2.1.       The Unversity of Ottawa is pleased to invite qualified proponents to submit a proposal for pre-clinical optical in-vivo imaging system (IVIS) to allow the evaluation of cell- and gene-based diseases & therapies using fluorescent, or luminescent tracing methods.

1.2.2        Scope of Project:

The requested optical in-vivo imaging system (IVIS) is a critical piece of infrastructure required to allow the evaluation of cell- and gene-based diseases & therapies using fluorescent, or luminescent tracing methods in small animals (e.g., mice, rats, guinea pigs, small rabbits, and zebra fishes) in-vivo, ex-vivo specimens (e.g., organs and tissue sections) and in vitro applications (in microplates, petri-dishes, etc.). The IVIS is a necessary tool in oncology, virology, neuroscience, biodistribution studies, treatment studies, cell tracking, and ex-vivo imaging.

The equipment will enable state-of-the-art approaches e.g., to visualize and track tumours & infections, diseases, or inflammation development, or follow biodistribution and pharmacokinetics studies, or therapy progression within living animals in a non-invasive manner. In particular it will be used for the development of human rodent disease models, assess dysfunction in models of disease, and the evaluation of cell- and gene-based therapies using fluorescent, or luminescent labelling/tracing methods.

1.3           General Description of Instrument/Product, System or Service:

The in-vivo imaging system is a non-invasive visualization tool to study living organisms or samples for research or diagnostic purposes. The two main principles used are bioluminescence and fluorescence imaging.

Bioluminescence imaging is based on the reaction between a reporter gene (luciferase) and its substrate (D-luciferin). The injected substrate reacts with the reporter gene and light is emitted. Fluorescence imaging, on the contrary, is based on excitation of a fluorophore which subsequently returns to its original energetic state producing photons with higher wavelengths.

Various bioluminescent reporters like a firefly luciferase and many fluorescent molecular reagents can be used to visualize and track tumours, disease or inflammation development, target molecules to nanoparticles or follow biodistribution and pharmacokinetics studies within living animals in a noninvasive manner.

1.4          Specifications and Standards:

The proposed system must be capable of performing in-vivo, ex-vivo and in-vitro imaging of small animals and detect fluorescent and bioluminescent sources.

Camera & Optics
The CCD camera operating temperature must be -90°C or lower.
To capture as much light as possible the lens aperture must allow f/stops being f/1.2 or lower (e.g., f/0.90) .
The peak quantum efficiency (QE) must be at least 80% within the range 500 – 700nm.
The native camera resolution must be at least 2000x2000 or higher with a pixel size of 15mm or lower (e.g., 8.0mm).
The camera must be a 16-bit CCD Camera with high optical density.
The proposed system must be calibrated to a NIST standard.
Field of view (FOV): the proposed system must be capable of imaging up to min 5 mice or 2 rats at a time as a standard across all modalities and allow macro imaging.

Illumination
The proposed system must include min 8 excitation channels from ~400 to ~800nm
The proposed system must include min 8 narrow bandpass emission filters from ~500 to ~900nm
The proposed system must allow visualizing several reporters simultaneously i.e., multi-spectral capabilities and spectral unmixing features for both bioluminescence and fluorescence imaging.

Hardware
The proposed system must have a fully software-controlled motorized system for lens, camera, and sample stage.

Animal Management
The proposed system must have a heated platform.
The proposed system must have an integrated gas anesthesia platform with an animal breather up to min 5 mice.
The proposed system must have a gas scavenging platform.

3D Imaging
The proposed system must provide 3D mapping of the target object for 3D source signal volume reconstruction allowing labelling and multimodal fusion.

Specifications listed in Appendix A – Technical Specifications Compliance Form of this RFP are the mandatory minimum requirements for the pre-clinical optical in-vivo imaging system (IVIS).

1.5           Should any equipment included in a proposal be superseded by a newer model during the period between the award of contract and the delivery date, the successful Proponent will be required to inform the University of Ottawa of the new specifications, the newer model pricing will remain the same as what was offered in the RFP. At its sole discretion, the University of Ottawa may opt to make changes from the list of equipment by altering, adding, or deducting some equipment.
